Cloning and Analysis of Differentially Expressed Genes from Peanut in Response to Chilling Stress

Abstract

Differences in gene expression between seeds of Huayu 44, a large type peanut line with chilling tolerance imbibed in 2℃ water and seeds of the same cultivar treated with 15℃ water (control), were studied using the GeneFishing technology. Two differentially expressed genes ACP20 and ACP2 were isolated. BLAST showed that they had high homology to the genes encoding oleosin and nifU-like protein, respectively. Expressions of both genes were studied by real time RT-PCR, and complete coding sequences of both genes were obtained by rapid amplification of cDNA ends. Preliminary biological functions of the two genes were further predicted. The study can help us to understand the molecular mechanism underlying resistance and facilitate breeding process.
