p38MAPK signaling pathways after activation of the astrocytes in neuropathic pain

Abstract

Objective:To investigate p38 mitogen-activated protein kinase (p38MAPK) signaling pathways after astrocytes were activated in neuropathic pain. Methods:20 SD rats were randomly divided into two groups:a model group of chronic constriction injury (CCI) in the sciatic nerve and a sham operation group. In every group, No. 4-5 lumbar spinal cord was taken to make paraffin slices at 1 day before operation and 1, 3, 7, 14 days after operation. The expression of p38MAPK was labeled by immunofluorescence method, and then the relationship between the nerve cells of the spinal cord and p38MAPK was detected with double immunolabeling technique. Results:The p38MAPK positive cells were increased in the number in the ipsilateral spinal dorsal horn of the operative limb in the CCI group. The mean fluorescence intensity of the p38MAPK was significantly increased compared with others, which reached the highest at 7 days after operation. The p38MAPK and micro-glia had a good consistence in the spinal dorsal horn in the CCI group. Conclusion:In the neuropathic pain, p38MAPK signaling pathways were activated, but the p38MAPK signal transduction pathways were not involved in pain signaling transduction of astrocytes.
