Expression of neuropeptide Y and leptin in hypophysis of type Ⅱ diabetic rats and the protective effects of sericin

Abstract
Objective:To investigate the effects of sericin on neusopeptide Y (NPY) and leptin expression in hypophysis of type II diabetic rats. Methods:Male SD rats were randomly divided into 4 groups:normal group, model group, sericin group and metformin group. The rats in the model group, sericin group and metformin group were given by continuous intraperitoneal injection of streptozotocin and blood glucose≥l6. 7 mmol/L was taken as the standard of modelformation. After successfully establishing diabetes model, the rats in the sericin group and metformin group were lavaged with sericin and metformin for 35 d. The blood glucose was detected. Immunohistochemical staining and RT-PCR were used to detect the expression of NPY and leptin and their mRNA in hypophysis . Results:Compared with diabetes model rats, the blood glucose, the expressions of NPY and leptin and their mRNA in the sericin group and metformin group decreased significantly; Moreover, there were no remarkable differences between two groups. Conclusion:Sericin can down-regulate the high expression of NPY and leptin in hypophysis and has protective effects on hypophysis injury of type fJ diabetes rats. Moreover, the effects of sericin on hypophysis injury of type II diabetes rats are similar with metformin.关键词
