Abstract
Objective To study the effects of ivabradine on serum HIF-la and VEGF in rabbits with acute myocardial infarction (AMI). Methods Twenty-one New Zealand white rabbits were randomly divided into four groups: sham operated group (S group) , myocardial-infarction group (M group) , bisoprolol-treated group (M + B group) and ivabradine-treated group (M+I group). The medication in M+B group started on the day of the model created, bisoprolol fumarate (1. 25 mg/d) was given for one week and then increased to 2. 5 mg/d for two weeks; In M+I group, ivabradine 17 mg/(kg ? d) by gavage for three weeks. Serum HIF-la and VEGF of rabbits in each group were detected-24 h before surgery and 24 h, three days, and one to three weeks after surgery-by ELISA method, and their heart rates, before and three weeks after treatment, were measured by ECG and analyzed. Results Three weeks after surgery, the heart rates of rabbits in M+B and M+I groups were slower than that in M group (F = 14. 97;q = 7.25,5. 97;P<0. 05) ,but that were not significant difference between M+B and M + I groups (P>0. 05). HIF-la and VEGF in M, M+B and M + I groups increased significantly compared to S group (F = 8. 41 — 48. 19,q = 3. 73 — 15. 48,P<0. 05). The levels of HIF-la and VEGF in M + B and M+I groups were lower than that in M group (q = 3. 97 — 8. 32, P<0. 05), but the difference of that between M+B and M+I was not significant (P>0. 05). There was a positive correlation between HIF-la and VEGF in each group (r = 0. 732,P<0. 01). Conclusion While reducing the heart rate, ivabradine can also reduce post-AMI level of HIF-la and VEGF in serum, its efficacy is similar to bisoprolol.关键词
