“泰山红”石榴高频再生体系构建
Establishment of High-Frequency in vitro Regeneration System for Taishanhong Pomegranate
摘要
Abstract
The tissue culture was conducted with stem segments of Taishanhong pomegranate shoots as explants. High - frequency in vitro regeneration system was established, which could lay foundation for genetic transformation and breeding. The best primary culture medium was MS + 1.5 mg/L BA +0. 3 mg/L IB A + 2 g/L PVP,and the rate of budding reached up to 85. 7% with the average shoot number of 4. 3. The optimal proliferation medium was B5 + 0.6 mg/L BA + 0.5 mg/L IBA + 0.3 mg/L AgNO3 + 0.2 mg/L GA3 + 100 ml/L CW(coconut water) + 2 g/L PVP,and the proliferation index reached 5. 8. The most suitable rooting media was B5 +1.0 mg/L IBA + 100 ml/L CW.and the rooting rate was 94. 0% and the survival rate of transplantation reached 89.0%.关键词
