Abstract

Objective To study the Magnetic Iron Oxide Nanoparticles as a carrier-mediated MDR1 asODN on human colorectal Carcinoma cells, and evaluate the targeting delivery effect. Method LS174T、HCT-8/ VCR cultured with PLL-DCIONP-asODN-FAM、 asODN-FAM in vitro were observed under fluorescence microscope. LS174T、 HCT-8/VCR cultured with PLL-DCIONP-asODN、 PLL-DCIONP in vitro were observed by Perle's blue staining. In addition, the change of magnetic singal intensity was measured by 1.5T MRI. Results The fluorescence intensity of transfected PLL-DCIONP-asODN-FAM in LS174T, HCT-8/VCR cells was significantly higher than asODN-FAM transfection group. The blue iron particles of transfected PLL-DCIONP-asODN in LS174T, HCT-8/VCR cells were significantly more than PLL-DCIONP transfection group. After transfaction by PLL-DCIONP-asODN the cells spin-spin relaxitions were shortened gradually compared with increasing labeled cells and showed a trend to attenuation. Conclusions PLL-DCIONP-asODN has high targeting delivery effect on HCT-8/VCR.
