The Cloning and the Sequence Analysis of Southern Xinjiang Strain ORF5 Gene of Porcine Reproductive and Respiratory Syndrome Virus

Abstract
[ Objective ] This project aims to grasp the characteristics of pandemic strain of the Southern Xinjiang PRRSV and provide a theoretical basis for the effective prevention and control of the virus. [ Method] In this study, eight complete sequences of 0RF5 genes of the South Xinjiang PRRSV were obtained by cloning and sequence analysis. [Result] The gene length of eight complete sequences of ORF5 genes of the Southern Xinjiang PRRSV were all 603 bp, nucleotide homology between them was 87.4% to 100.0% ; The nucleotide homology of ORF5 genes were 60. 9% to 63. 5% , 88. 4% to 89. 9% , 88. 9% to 94. 7% , and 87.4% to 98. 7% , respectively, compared with LV of Europe type representative strain, ATCC VR -2332 of American type representative strain, CH - la of China type representative strain, and JXA1 of the HP -PRRS 9China type representative strains. And compared with attenuated vaccine strains used wildly in China pig farms, their nucleotide homology was 87. 4% to 94.7% . The amino acids homology of ORF5 genes compared with the American type representative strain and Europe type strain was 84. 5% to 98. 0% and 50. 0% to 59. 5% . [ Conclusion] The five ORF5 genes of the Southern Xinjiang PRRSV all belong to the American type subgroup Ⅱ strains. The molecular epidemiology investigation result of the Southern Xinjiang PRRSV has laid the foundation for the further adjustment and guidance in the choice of vaccine for Southern Xinjiang PRRS.关键词
