Effects of simvastatin on the proliferation and osteoblastic differentiation of human dental pulp stem cells

Abstract
Objective To study the effects of simvastatin on the proliferation and osteoblastic differentiation of human dental pulp stem cells (DPSCs). Methods The third passage human DPSCs were cultured in conditioned mineralization medium with different concentrations of simvastatin. Proliferation ability was detected by MTT method.Alkaline phosphatase activity was measured with ALP kit. Results Proliferation of human DPSCs was suppressed in all treatment groups with different concentrations of simvastatin(10-5mol/L,10-6mol/L,1×10-7mol/L,1×10-8mol/L). The maximum effect on the proliferation of DPSCs was observed at the concentration of 1×10~5mol/L.Osteoblastic differentiation of human DPSCs was increased in treatment group with different concentrations of simvastatin(10-6mol/L, 1 ×10 -7mol/L,1 ×10-8mol/L). The maximum effect on the activity of ALP was observed at the concentration of 1×10-7mol/L. Conclusion Proliferation of human DPSCs was suppressed by simvastatin.Optimal concentration of simvastatin can improve the osteoblastic differentiation of human DPSCs.关键词
