Abstract
Objective To investigate the expression of MACC l,HGF,and C-met in endometrial carcinoma and their clinical significance. Methods The expressions of MACC1 ,HGF and C-met in 25 specimens of normal endometrium tissues, 25 specimens of simple hyperplasia endometrium tissues,25 specimens of atypical hyperplasia endo -metrium tissues , and 39 specimens of endometrial carcinoma were measured by immunohistochemistry and reverse transcription-poly-merase chain reaction (RT- PCR). The correlations of the expressions of MACC1, HGF and C-met to the clinicopathologic characteristics of endometrial carcinoma were analyzed, and the correlations between the expressions of the 3 proteins were also evaluated. Results The positivity rates of MACC1 ,HGF and C-met proteins were 64. 1% ,61. 5% ,and 69. 2% in endometrial carcinoma with relative mRNA expressions of 0. 216 ±0. 062,0. 134 ±0. 054,0. 172 ±0. 044,showing significant differences from those in normal endometrium tissues, simple hyperplasia endometrium tissues, and atypical hyperplasia endometrium tissues (P<0. 05). In endometrial carcinoma,the up-regulation of MACC 1,HGF and C-met expressions were associated with advanced FIGO stage,muscle layer infiltration and lymph node metastasis ( P <0. 05). MACC1 expression was positively correlated to HGF (r = 0. 525, P = 0. 000) and C-met expressions ( r = 0. 393, P = 0. 008) , and the latter two was also positively correlated (r = 0. 334, P = 0. 027). Conclusion MACC1 may serve as a potential bio-marker for advanced endometrial carcinoma. Deregulation of MACC1, HGF and C-met proteins may synergistically participate in the malignant progression of endometrial carcinoma.关键词
