摘要
Abstract
Objective To explore the effect of hTERTC27 overexpression on the proliferation and apoptosis of nasopharyngeal carcinoma C666-1 cell line. Methods pcDNA3-hTERTC27 was transfected into nasopharyngeal carcinoma C666-1 cells with lipofectamine. The expression of hTERTC27 and Bcl-2 were detected by Western blot. The proliferation of cells was detected by MTT assay and colony formation assay. The apoptosis of cells was detected by TUNEL assay. Results C666-1 cells expressed high level of hTERTC27 after 24h transfection. Over expression of hTERTC27 obviously suppressed the cell prolifera-tion(P<0. 01) ,downregulated the colony formation rate(P<0. 01) Increased the TUNEL positive cells (P<0. 01) ,and downregulated the expression level of Bcl-2 protein(P<0. 01). Conclusion Overexpression of hTERTC27 can effectively inhibit proliferation and promote apoptosis of nasopharyngeal carcinoma cells, which is possibly due to the down regulation of Bcl-2 protein.关键词
