Extraction of genomic DNA and optimization of ISSR reaction system with orthogonal design in Glyptostrobus pensilis
摘要
Abstract
The improved CTAB method was used for the extraction of genomic DNA in different populations of Glyptostrobus pensilis, and a satisfactory ISSR reaction system was also established by orthogonal design. The result showed that the improved CTAB method was suitable for the extraction of DNA, and the DNA extracted by this method had higher yield, better quality, and could be used for ISSR-PCR reaction. A number of factors, which affected the results of ISSR - PCR reaction, such as quantity of template DNA, primer, concentration of dNPT and Taq DNA polymerase were studied.关键词
