Abstract

AIM: To investigate the effect of FasL expression in bone marrow mesenchymal stem cells (BMMSCs) on CD4+ T-lymphocyte apoptosis during osteoporosis (OP) under the condition of estrogen deficiency. METHODS: An animal model of OP was established by ovariectomy (OVX, bilateral ovarian resection) in 8-week-old healthy female mice. T-lymphocytes were divided into OVX group, sham group and estradiol-stimulated group, and co-cultured with the BMMSCs from the mice. The comparison of CD4+T-lymphocyte apoptosis among the 3 groups was performed. The level of FasL expression was detected by Western blotting and real-time PCR. RESULTS: The apoptosis of CD4 + T-lymphocytes was significantly higher in sham group than that in OVX group. The expression of FasL at mRNA and protein levels was lower in OVX group than that in sham group, and was increased by estrogen in a concentration-dependent manner. CONCLUSION; Expression of FasL is regulated by estrogen in BMMSCs. The change of FasL affects apoptosis of CD4+ T-lymphocytes, which may be an important mechanism in the pathogenesis of postmenopausal osteoporosis.
