Abstract
Objective To evaluate the pharmacokinetics of multiple -dose of cefazedone sodium injection in Chinese healthy subjects. Methods An open, mulliple - dose intravenous study was conducted. Twelve subjects were administrated intravenously 2 g cefazedone sodium within 45 min, Q12 h for 7 days. The blood and urine samples were collected at day 1 and day 7 respectively. At day 4 - 6, only collected blood sample before infusion. The concentrations of cefazedone in serum and urine were determinated by HPLC method. Results The main pharmacokinetics parameters at day 1 and day 7 were as follows: Cmax were (242. 18 ± 44.02) and (228. 82 ± 40. 71) mg ? L-1;tmax were (0. 75 ±0.00), (0.75 ±0.00) h; t1/2(} were (1.68 ±0.20) and (1.61 ±0. 16) h; AUC0-1,were (529.48 ±98.63) and (441.93 ±80.78) mg ? H ? L-1; AUC0-∞ were (535.69 ±98.63) and (446.51 ±82.41) mg ? H ? L-1; Vd were (9. 44 ± 2. 68) and (10.63 ± 1. 61) L; CL were (3. 86 ± 0. 76) and (4. 62 ±0. 87) L ? H-1 ; respectively. Urinary recovery rates were (78. 36 ± 10. 82) and (78. 54 ± 13. 62) % , respectively. Conclusion The results showed that the main pharmacokinetics parameters of cefazedone sodium at day 1 and day 7 were similar. There was no accumulation in human body.关键词
