Abstract
Objective To determine the concentration of meropenem in human serum and investigate its pharmacokineties in Chinese elder patients in ICU. Methods A single 0.5 -1.0 g dose of meropenem was given to 10 elder patients in ICU by infusion administration route. The age of patients exceeded 65 year and the drug was administrated routinely. Blood samples were proceeded by protein precipitation and the serum concentrations of meropenem were determined by HPLC method, which was built in this study. Data were analyzed by the WinNonline version 5.2 software. Results The major pharmacokineties parameters were; Cmax/Dose = (55.74±18.65) μg ? mL-1.g-1; t1/2 = (2. 62 ± 1. 67)h; CL = (39.34±5.94) L? h-1; X2 = (0.31 ±0. 12)h1; V=(28.97± 13.42) L: AUC0-1, = (97.22 ±45.49) μg ? H ? mL-1. Conclusion The method described in this report was of high sensitivety and selectivity for accurate determination of the serum concentration of meropenum in human. The pharmacokineties results obtained from this study showed that the pharmacokineties of metopenum in elder patients was significantly different from adult patients.关键词
