Methionine Sulfoxide Reductase A in Colon Cancer Tissues and Its Significance

Abstract
Objective Objective Exploring the Methionine Sulfoxide Reductase A ( MsrA) mRNA expression and intraeellular reactive oxygen species (ROS) levels in colon cancer cell, and exploring the MsrA mRNA expression in different histological grading of colon cancer. Method Detecting MsrA mRNA expression in colon cancer cell and in different histological grading of colon career by reverse transcription-polymerase chain reaction (RT-PCR) ; detecting the reactive oxygen species levels in colon cancer cell by 2'-7'-dichlorofluorescein diacetate salt (DCFH-DA) . Result The fluorescence of ROS in colon cancer cell was 350. 2 ±6. 3; it was significantly higher than that in normal colon cells 134.3 ±15.6, the difference was statistically significant ( P < 0. 05) . MsrA mRNA expression levels in colon cancer cells was 1. 27 ±0. 32, it was significantly lower than that in normal colon cells 0. 62 ± 0. 27 , the difference was statistically significant ( P < 0. 05 ) . MsrA mRNA expression in histological grade Ⅰ of colon cancer cells was 0. 93 ±0. 26, it was significantly higher than that in the histological grade Ⅱ of colon cancer cells 0. 35 ±0. 29 or in histological grade H of colon cancer cells 0. 32 ± 0. 20, both of the differences were statistically significance (P < 0. 05) , but MsrA mRNA expression were no significant difference between Histological grade Ⅱ co- Ion cancer cells and histological grade Ⅲ colon cancer cells (P > 0. 05) . Conclusion MsrA may be involved in colon cancer differentiation by regulating the level of intracellular reactive oxygen species .关键词
