Abstract

Objective;To provide a basis for the pathogenesis of preeclampsia by investigating whether syncytiotrophoblast microparticles (STBM) could cause the maternal vascular endothelial dysfunction. Methods: Placentas were obtained after elective cesarean section from four uncomplicated pregnancies from June 2009 to Oct. 2009 in the Sixth Affiliated Hospital of Shanghai Jiaotong University. Different concentrations of STBM and RBC ghost particles were cocultured with human umbilical vein endothelial cells. Proliferation and apoptosis rates of human umbilical vein endothelial cells were measured by MTT and flow cytometry. Results; ( 1 ) When human umbilical vein endothelial cells were incubated together with STBM preparation, the cell monolayer was disrupted,leading to cell detachment. (2)After48h of incubation,large amounts of human umbilical vein endothelial cells detached and the number of cells were significantly reduced. STBM at 30,90 and 150mg/L respectively caused an average of (55. 58 ± 2. 40) % , (67.94±1. 93) % and (86. 37 ± 1. 59) % inhibition of proliferation. There were significant differences among the groups( P<0. 01 ). STBM preparations inhibited human umbilical vein endothelial cells proliferation in a dose-dependent manner. (3 ) After 24h, the percentage of apoptosis was (17.39±1.11)% ,(22. 13±1. 02)% , (30. 42±1. 98)% for STBM at 30,90,150mg/L,respectively. With the raised concentration of STBM,apoptosis rate of human umbilical vein endothelial cells significantly increased (P<0. 01). Conclusion: STBM inhibits the proliferation and induces cells apoptosis of human umbilical vein endothelial cells, which may contribute to the endothelial dysfunction underlying the maternal syndrome of preeclampsia.
