Abstract
Objective: Study the expression and changes of costimulatory molecule CTLA-4, B7-1 and B7-2 in experimental autoimmune myocarditis ( EAM) and explore the pathogenesis of EAM. Methods: EAM model was established by injecting porcine cardiac myosin into Lewis rats(experimental group, n =30) ,and then the 30 rats were randomly divided into three subgroups in accordance with 14 d, 28 d, 56 d after the first immunization . HE staining was used to observe the change of histopathological characteristics. Im-munohistochemical staining and real-time fluorescence quantitative PCR detection was used to analysis the expression of CTLA-4, CD80, CD86. Results:There were varying degrees of myocarditis change in experimental group; CTLA-4, CD80, CD86 mainly expressed in cardiac cells. After the first immunization, expression of proteins increased gradually from 14 d to 28 d, but decreased in 56 d. Real-time PCR: compared with normal group, the expression of CD80, CD86 mRNA were higher in experimental group ( P < 0. 05 ) . Conclusion: The expression of CD80, CD86 and CTLA-4 promotes heart tissue inflammation, and may closely related to the occurrence and development of EAM.关键词
