Changes and roles of TNF-α, IL-1β, IL-6 and IL-10 in acute lung injury induced by H9N2 subtype swine influenza virus in mice

Abstract

Objective To explore the changes and roles of inflammatory factors in acute lung injury induced by swine influenza virus H9N2 in mice. Methods Eighty 6-8 week old female Balb/c mice were divided into experimental and control groups (n =40). The mice of experimental group were infected with swine influenza virus (H9N2) by nose dropping. Lung tissue specimens of the mice were taken on day 2, 4, 6, 10 and 14 after infection, and the concentrations of TNF-α, IL-1β, IL-6 and IL-10 in the tissue homogenate were determined. Results Concentrations of TNF-a, IL-1β, IL-6 and IL-10 in the lung tissue homogenate of infected mice were significantly higher than that in the control group at different time points (P < 0. 05 ) . TNF-α and IL-1β were tended towards normal, but IL-6 and IL-10 were still at high levels after 14 days. Conclusions Inflammatory factors play a major role in acute lung injury induced by H9N2 subtype swine influenza virus in mice. TNF-a and IL-1β display a proinflammatory effect, while IL-10 is likely to play an anti-inflammatory role, similar to that of IL-6.
