超声影像分析在高脂血症金黄地鼠模型构建中的动态监测
Establishment and dynamic ultrasonic imaging analysis of hamster models of hyperlipidemia
摘要
Abstract
Objective To study the changes of liver, aortic wall and heart function in hyperlipide-mic hamster at different time points by ultrasonic imaging, and evaluate the value of ultrasonic diagnosis in hamster model of hyperlipidemia. Methods Thirty-two Syrian golden hamsters were randomly divided into two groups: the control group ( n = 8 ) was fed with normal chow and the model group ( n = 8) fed with high fat diet. The levels of serum lipids were analyzed after 0,3,6,9, 14, 16, 20, 24, 40 weeks using commercial kits. Hamsters from each group were examined by ultrasound and then sacrificed for pathological examination at the 20th or 40th weeks. Results The levels of serum TC, TG, HDL-C and LDL-C were remarkably increased in hamsters fed with high fat diet and maintained significantly above the normal levels throughout the study course ( P < 0. 05 ). Compared with the control group, hamsters fed with high fat diet for 20 weeks had increased intima-media thickness (IMT) (P <0. 000 1) and the IMT were significantly increased when the dietary intervention period was extended to 40 weeks (P < 0. 000 1). Fatty liver and aortic arch atherosclerotic lesions in the hyperlipidemic hamsters fed with high fat diet for 20 weeks were revealed by both ultrasonic imaging and pathological examination, and were further developed and aggravated during the prolonged 20 weeks. Conclusions Ultrasonic imaging can be used to measure the increased aortic arch intima-media thickness of hamsters, and can also be used to monitor the dynamic development of fatty liver and atherosclerotic changes of the aortic arch wall in hyperlipidemic hamsters.关键词
