Abstract

Objective :To observe the effect of icariin on s-nitrosoglutathione reductase (GSNOR) and Bcl-2 in diabetic rats testis. Methods: Sprague-Dawley rats were induced by an injection of streptozotocin. Four weeks after injection of STZ, the rats were randomly divided into 5 groups: a normal group, a control group, a low dose group (30 mg/kg, ig), a middle dose group (70 mg/kg, ig) and a high dose group (100 mg/kg, ig). The blood of the heart was tested for testosterone and estradiol. Im-munohistochemical technique was used to detect the expression of GSNOR and Bcl-2. Results: Compared with control group, the ratio of testosterone and estradiol in the middle dose group were increased, and the fasting food glucose was descend evidently between the control group and the middle dose group. Numerous pathological changes (decreased spermatogenic cell, distortion seminiferous tubule) were lessening in diabetic rats after receiving ICA. Immunohistochemistry showed that there were higher GSNOR protein expressions in the low dose group and middle dose group. A high expression of Bcl-2 protein was observed in Leydig cells. Significant differences existed in the control group, the low dose group and the middle dose group. Conclusion: Icariin evidently relieves testicular damage in diabetic rats by improving the ratio of testosterone and estradiol,reducing the level of FFG and increased the expression of GSNOR and Bcl-2 protein.
