Regulation of 14-3-3 in the First Mitotic Cell Cycle in One-cell Stage Mouse Fertilized Eggs

Abstract
Objective To study the effects of 14-3-3 proteins in regulation of the first mitotic cell cycle in one-cell stage mouse fertilized eggs. Methods 14-3-3 isoform in the mouse fertilized eggs was identified by RT-PCR. 14-3-3 siRNA was introduced to G1 phase fertilized eggs by microinjection to study the cleavage rate, morphology and MPF activity. Results 14-3-3 ε was identified in one-cell stage of mouse fertilized eggs. Compared with the control group,the cleavage rate in pSUPER-14-3-3ε siRNA injection group was significantly decreased, mitosis was delayed and more abnormal morphology eggs were observed. Moreover, the maximal value of MPF activity was significantly decreased. Conclusion 14-3-3 proteins play critical roles in the first mitotic cell cycle in mouse fertilized eggs.关键词
