Abstract

Objective To investigate the effects of 17-AAG on VEGF and STAT3 expressions in human breast cancer MCF-7 cells. Methods MCF-7 cells were cultured in DMEM medium; MTT method was performed to evaluate the inhibitory effects of 17-AAG with different time and different doses on proliferation of MCF-7 cells;the genes and proteins expressions of VEGF and STAT3 were determined by RT-PCR and Western blot analysis. Results With treatment of 0.165-10.000 mg/L 17-AAG for 24 hours and 48 hours, significantly inhibition of the MCF-7 cells proliferation was observed in both time- and dose-dependent manner. The assay of RT-PCR and Western blot showed that the genes and proteins expressions of VEGF and STAT3 in MCF-7 cells induced by 17-AAG were significantly down-regulated in a dose-dependent manner when the cells were treated with 1.0,2.0,3.0,5.0 mg/L 17-AAG for 48 hours. Conclusion 17-AAG can inhibit the cell proliferation and down regulate the genes and proteins expressions of VEGF and STAT3 of human breast cancer MCF-7 cells in vitro.
