Abstract
Objective To study the the necrotic vascular smooth muscle cells, and to explore its possible mechanism of action of the surrounding normal cell proliferation and inflammatory cytokine secretion. Methods Cells were cultured in low oxygen serum free and glucose deprivation medium to establish cell necrosis model. Necrotic cell culture supernatant was collected to interfere normal vascular smooth muscle cell. Experiment groups were set as necrosis supernatant group, IL-1 group, necrosis supernatant of IL-1RA group and control group, MTT assay of cell proliferation; RT-PCR was used to detect the groups of MCP-1, IL-6, IL-8 mRNA expression; ELISA to detect the MCP-1, IL-6,IL-8 secretion; Western blot to detect the activation of NF-κB. Results NCS group and IL-1 cell proliferation were significantly higher than that in the control group (P<0.01), NCS Group IL-1 beta cell proliferation was significantly lower than that of the NCS group and IL-1 group (P<0.01); NCS group and IL-1 β, MCP-1, IL-6, IL-8 mRNA expression were significantly higher than those in the control group (P<0.05) the NCS of IL-1RA groups MCP-1, IL-6, IL-8 mRNA expression was significantly lower than the NCS group and IL-1 group (P<0.05); NCS group and IL-1 β MCP-1, IL-6, IL-8 protein secretion, significantly higher than the control group (P<0.05), the NCS of IL-1RA groups MCP-1, IL-6, IL-8 protein secretion was significantly lower than the NCS group and IL-1 group (P<0.05), the NCS group and IL -1 NF-κB activity was significantly higher than that of the control group (P<0.05), compared with the NCS group and IL-1 group NCS IL-1RA group of NF-κB activity was significantly decreased (P<0.05). Conclusion The necrosis vascular smooth muscle cells can promote the proliferation of normal cells, induction of NF-κB activation and inflammatory cytokine release, and this role may be related to the massive release of IL-1.关键词
