Abstract
Objective To explore the expression patterns of stem cell transcription factor SOX2 and its significance in the early diagnosis and prognosis of human esophageal squamous cell carcinoma. Methods The expressions of SOX2 in esophageal squamous cancer cell lines KYSE70, KYSE140 and KYSE450 were characterized with real-time PCR, Western blotting and immunocytochemistry. The expression of SOX2 in 153 esophageal squamous cell carcinoma specimens was detected with immunohistochemistry. The correlation between the clinicopathological features and SOX2 expression in esophageal squamous cancer was analyzed with Crosstabs analysis, while survival curve regarding SOX2 expression was observed using the Kaplan-Meier method. Results SOX2 was variably expressed in the three esophageal squamous cell carcinoma cell lines. Immunohistochemically, SOX2 positive immunostaining was observed in the cancer cell nuclei. Among the 153 esophageal squamous cell carcinoma specimens, 98(64.1%) were either negative or weakly positive for SOX2; 36 (23.5%) were moderately positive while 19(12. 4%) were strongly positive. Crosstabs analysis showed that higher SOX2 expression was significantly associated with higher histological grade and poorer overall survival. SOX2 expression level was an independent factor affecting the prognosis of esophageal squamous cell carcinoma and the relative risk was 1. 976. Conclusion Higher expression level of stem cell transcription factor SOX2 in esophageal squamous cell carcinoma is significantly associated with higher histological grade and poorer overall survival of patients, which makes SOX2 an independent prognostic factor of survival.关键词
