Abstract

Objective: To observe the effect of tetrame-thylpyrazine (TMP) on cell proliferation and cell cycle phase of human cervical carcinoma HeLa cell line and to elucidate its relationship with CXCR4 expression. Methods :HeLa cells were treated with TMP for 48 hours. Methylthiazolyl tetrazolium assay was performed to reveal the inhibitory effect on cell proliferation, and flow cytometry was used to determine the cell cycle of the HeLa cell line. Western blot and immunofluorescence were adopted to evaluate the expression of CXCR4 on the HeLa cells. Results: We found that TMP remarkably inhibited cell viability and growth of HeLa cells by MTT (P <0. 01). Flow cytometry demonstrated that TMP increased the rate of G0/G1 of HeLa cell cycle (P <0. 01). Western blot and immunofluorescence results showed that TMP down-regulated the expression of CXCR4 in HeLa cells. Conclusion: TMP can inhibit the viability and growth of human cervical carcinoma HeLa cell line. Our results indicate that TMP-mediated treatment involve inhibiting the expression of chemokine CXCR4 in HeLa cells.
