The expression of P2Y2 and its effect on intracellular Ca2+ in hepatocellular carcinoma cell

Abstract
Objective To investigate the expression of P2Y2 receptor in hepatocellular carcinoma (HCC) cells and its effects on intracellular Ca of HCC cells. Methods The quantitative real - time RT - PCR was applied to assess the expression of P2Y2 receptor mRNA in normal hepatic cells (LO2) and HCC cell lines , including HepG2 and Bel -7402. The intracellular Ca signal after stimulation by P2Y receptor special agonist ATP was examined by confocal laser scanning microscope. Results The mRNA expression of P2Y2 in HCC cells were significantly higher than that in normal hepatic cells (P < 0. 05 ). Compared to the normal hepatic cells , significantly more prominent increase of intracellular Ca in HCC cells were induced by ATP (P < 0. 05 ) , which was reversed by Suramin. Conclusion P2Y2 receptor is up - regulated in HCC cells and involved in the regulation of intracellular Ca , suggesting important role in the develop -ment and progression of HCC.关键词
