Abstract

According to the gene sequences of Marteilia refringens in GenBank, one pair of specific primers were designed for amplifying the specific fragments of M. refringens. The reaction parameters were optimized to develop the two-temperature PCR method for detection of M. refringens. The specific DNA fragment with the length of 266 bp was amplified from M. refringens, but not from other pathogens such as Haplosporidium sp. , Perkinsus sp. , Vibrio parahaemolyticus, Vibrio alginolpicus, and Vibrio fluvialis. The sensitivity results showed that as little as lfg DNA plasmid of M. refringens could be detected. 256 Shellfish samples from Fujian were detected by this two-temperature PCR. As a result, 12 razor clam samples were positive. It suggested that M. refringens existed in the cultivated shellfish in Fujian and this two-temperature PCR could be used as a sensitive tool to detect M. refringens in clinical samples.
