人胃癌细胞原位移植转移裸鼠原发灶及转移灶中锌指蛋白139的表达及其与肿瘤侵袭转移的关系研究
Expression of ZNF139 and Its Relationship with Invasion and Metastasis in Primay Tumors and Metastases of Orthotopic Implantation Nude Mice

Abstract
Objective To investigate the expression of zinc finger protein 139 ( ZNF139 ) and its relationship with invasion and metastasis in the primary tumor and metastases of orthotopic implantation nude mice.Methods Orthotopic implantation nude mice with human gastric carcinoma cells were established.Then reverse transcription - polymerase chain reaction ( RT - PCR ) and immunohistochemical method ( S - P ) were used to detect the expression of ZNF139, MMP - 2 and MMP - 7 in orthotopic transplantation tumor, liver metastases, spleen metastasis, peritoneal metastasis and lymph node metastasis.Results The tumor formation rate was 93.3% ( 28/30 ), and hepatic metastases were 75.0% ( 21/28 ), spleen metastases were 67.9% ( 19/28 ), peritoneal metastases were 85.7% ( 24/28 ) and lymph node metastases were 39.3% ( 11/28 ) .The mRNA expression and protein positive expression rate of ZNF139 , MMP - 2 and MMP - 7 in liver metastases, peritoneal metastases and lymph node metastases were significantly higher than in primary tumor ( P < 0.05 ) .The expression of ZNF139 mRNA was positively correlated with the expression of MMP - 2 mRNA and MMP - 7 mRNA both in primary tumor and metastases ( P < 0.05 ).Conclusion Models of orthotopic implantation nude mice can well simulate the invasion and transfer process.Invasion and metastasis are stronger in metastases than in primary tumor, and ZNF139 may be involved in this process by promoting the expression of MMP -2 and MMP -7.关键词
