Isolation and molecular typing of methicillin-resistant Staphylococcus aureus from swine and human in Shanghai

摘要

Abstract

The objective of this study was to investigate the prevalence and characteristic of methicillin-resistant Staphylococcus aureus (MRSA) isolated from swine and human in Shanghai. A total of 170 swine nasal swab samples were collected from three large-scale swine farms and one pig slaughterhouse. A total of 120 sputum and throat swab samples of human origin were collected from the patients in one hospital. MRSA were isolated and identified by using 75 g/L sodium chloride broth and CHROM agar and duplex PCR(nuc and mecA). The MRSA isolates were characterized by staphylococcal chromosomal cassette(SCC) mec typing,multilocus sequence typing (MLST) and spa typing. Among 83 swine S. aureus isolates, 45 strains were resistant and among 52 human S. aureus isolates, 15 were resistant. All MRSA isolates of swine origin were determined to be ST9-SCC mec IVb,and characterized as spa type t899(n = 37) and t2922 (n = 2). Fourteen of the 15 human MRSA isolates were determined to be ST5-SCC mec II -t002,and one was characterized as ST88(MLST)-tl0775(a novel spa type). The results showed that swine MRSA were ST9-t899-SCCmec IVb,and human MRSA were ST5-t002-SCC mec II in Shanghai. There were no signs of MRSA transmission between human and pig.
