摘要

Abstract

Objective To examine the effect of high glucose on the apoptosis and proliferation of vascular smooth muscle cells ( VSMCs) and the change of apoptosis-related gene bcl-2 in the process. Methods VSMCs were isolated by the explant culture method and cultured in Dulbecco s modified Eagle's medium (DMEM) containing 10 % serum in the presence of 5 mmol/L glucose (normal glucose group) , 25 mmol/Lglucose (high glucose group) , or 5 mmol/L glucose plus 20 mmol/L mannitol ( mannitol group) . Real-time PCR and Western blotting were used to detect the bcl-2 expression in the three groups. MTT assay was employed to determine the proliferation of VSMCs and flow cytometry to analyze the apoptosis of VSMCs . Results The expression level of bcl-2 was significantly increased in VSMCs treated with high glucose compared with those cultured in normal glucose. MTT showed that cell proliferation was enhanced in high glucose group relative to normal glucose group. Flow cytometry showed that cell apoptosis was decreased in high glucose group when compared with the normal glucose group . Mannitol, used as an osmotic control, had no effect on cell apoptosis. Conclusion High glucose may contribute to the proliferation of VSMCs and eventually the occurrence of diabetic vasculopathy by stimulating the bcl-2 activity and inhibiting the cell apoptosis.
