Optimizing SRAP Reaction System for Abelmoschus esculentus Linn.

Abstract
Through optimization of the concentrations of Taq DNA polymerase,dNTP,primers and DNA template,SRAP-PCR of Abelmoschus esculentus Linn.was established.The results indicated that,in a 20 μL reaction mixture,the optimal concentration of genomic DNA template was 75 ng; that of the primers,0.4 mmol · L-1 ; that of dNTP,0.75 mmol · L-1 ; that of Taq DNA,polymerase,1.5 U; and that of MgCl2,2.0 mmol · L-1.关键词
