Effects of local injection of insulin-like growth factor-1 on the expression of integrin αV,β3 in periodontal tissues of orthodontic tooth in rats

Abstract
Objective To study the effects of recombinant human insulin-like growth factor (rhIGF-I) on integrin αV,β3 in the periodontal tissue of orthodontics tooth in rats.Methods The first maxillary molars of the rats were moved medially by an elastic string which delivered a force of 50 g.Thirty SD rats were randomly divided into six groups,which were injected with 0.1 mL rhIGF-1 of 0 (control group),1,2,4,8,16 μg/mL in the buccal submucosal area of the molar per triduum.Ten days later,all rats were killed and the expressions of integrin αV,β3 in periodontal tissue were deteceted in situ hybridization.Results Integrin αV,β3 of control group expressed lowly in periodontal tissues.The 2 μg/mL and 4 μg/mL groups showed a significantly increasing expression of integrin αV,β3 in the compression sides compared with the control group (P ＜0.01).At the level of 8 μg/mL and 16 μg/mL,the expressions reduced gradually,but showed significantly differences compared with the control group (P ＜ 0.05).The expressions of integrin αV,β3 in the tension sides increased gradually with the concentrations of rhIGF-1 increased from 2 μg/mL to 8 μg/mL.At the level 16 μg/mL,the expression of integrin αV,β3 decreased slightly,but still significantly higher than that in the control group(P ＜ 0.01).Conclusion Exogenous rhIGF-1 accelerate the periodontal tissue remodeling by enhancing the expression of integrin αV,β3 in periodontal tissue.关键词
