川芎嗪对角膜基质细胞中趋化因子受体CXCR4的影响
Effects of tetramethylpyrazine on chemokine receptor CXCR4 of rat corneal stromal cells

Abstract
Objective To investigate the effects of tetramethylpyrazine ( TMP) on rat corneal stromal cells and the correlation between them. Methods Rat corneal stromal cells from 8 to 10 weeks SD rats were isolated and cultured . CXCR4 expression was measured with cell immunofluorescence method . Comeal stromal cells were treated with TMP for 48 h. Methylthiazolyl tetrazolium (MTT) assay was performed to assess cell proliferation , while reverse transcription PCR (RT -PCR) and Western blot were adopted to evaluate CXCR4 expression. Results The viability and growth of rat cor-neal stromal cells were significantly inhibited with ( P < 0. 05) ; so was the mRNA and protein expression of CXCR 4 ( P < 0. 05). Moreover, TMP significantly down - regulated the expression of CXCR4 in rat corneal stromal cells in a dose - dependent manner. Conclusion TMP remarkably inhibits the viability and growth of rat corneal stromal cells by down -regulating chemokine receptor CXCR4.关键词
