Abstract

In order to improve the degradability of the a preserved PBS-degrading strain,five different mutagenic methods were compared.The strain DSCF-8 which could degrade PBS film was chosen as the starting strain for mutagenesis.By analyzing the morphological features and mitochondrial cytb sequence,strain DSCF-8 was identified as Fusarium oxysporum.It was indicated that the effects of combined mutation were stronger than that of single mutation,and generation rate of efficient mutant strains reached 4.55% and 1.30%,respectively.However,the transmissibility of those mutant strains were not stable after subculturing.Mutation induced by using single mutagen diethyl sulfate was much more effective than by ultraviolet and microwave,and the mutant strain D8B14 obtained was stable after subculturing.The optimal conditions were suggested as 5% diethyl sulfate with a trea ting time of 40 min.The degradation rate of mutant strain D8B14 was 34.49% higher than that of the starting strain,and the enzyme activity was improved by 23.86%.In addition,the degradability of this mutant strain was much higher at 35℃,and the pH tolerance became much stronger.
