Determination of the total polyphenol in Anhua dark-tea by Folin-Ciocalteu colorimetry

Abstract
Objective To establish a method for determination of the content of total polyphenols in Anhua dark-tea.Methods According to GB/T 8313-2008,the total polyphenol content in Anhua dark-tea was analyzed by spectrophotometry with Folin-Ciocalteu reagent and Gallic acid used as standards.The optimal determination conditions could be calculated according to the absorption at 765 nm by reacting 5 mL 0.2 mol/L Folin-Ciocalteu with 4 mL 7.5% Na2CO3 for an hour at 25 ℃.Results In this method,the concentration of Gallic acid showed a good linearity with absorbance at the range of 0.49 to 4.9 μg/mL,the average recovery was 98.0%,and RSD was 1.4%.The total polyphenols content in Anhua dark-tea was 2.49% and RSD was 1.3%.Conclusion The method was accurate and reliable for the determination of total polyphenol in Anhua dark-tea.关键词
