Abstract
Objective:To investigate the effect of HMGB1 on the expression of TGF-β1 of cadiac fibroblast induced by high glucose.Methods:The cardiac fibroblasts were isolated from 3 w C57/BL6 mice.The identification of cardiac fibroblasts was evaluated by immunocytochemical staining with vimentin.The cardiac fibroblasts were treated by high glucose for different times(0,6,12,24,48 h).After transfected with siR-NA,the cardiac fibroblasts were treated by high glucose for 6h.The mRNA levels of HMGB1 and TGF-β1 of cardiac fibroblasts were determinated by RT-PCR.Results:The cultured cells were in accordance with characteristics of cardiac fibrolast.The mRNA levels of TGF-β1 of cardiac fibroblasts were inhibited by HMGB1 siRNA after treated by high glucose for 6 h.Conclusion:HMGB1 may contribute to modulate the expression of TGF-β1 of cardiac fibroblasts induced by high glucose.关键词
