摘要

Abstract

Objective To explore the effects of clonazepam pretreatment on expression of γ-amino-nbutyric acid receptor γ2 (GABAA R γ2) subunit of hippocampus in epileptic rats.Methods Sixty healthy male SD rats were randomly divided into the following groups:sham operated control group,epilepsy group,and pretreatment group; epilepsy group was then divided into 6 h,12 h,1 d,3 d,7 d,15 d and 30 d subgroups; pretreatment group was then divided into 6 h,12 h,1 d pretreatment subgroups and pseudo-pretreatment subgroup.The pretreatment group was given clonazepam 6 mg/(kg/d) in twice by intragastric for 5 d.Then the models of epilepsy in epilepsy group and pretreatment group were established by injection of kainic acid.The expression of GABAAR γ2 in hippocampus was assayed at various time intervals by immunohistochemistry.Results Compared with the sham operated control group,GABAA R γ2 expression of CA1 zone since 1 d after seizure and CA3 zone at all time points after seizure in the epilepsy group were significantly descended (P < 0.05-0.01).Compared with the epilepsy corresponding time subgroups,GABAAR γ2 subunit expressions of CA3 zone in 6 h,12 h pretreatment subgroups and CA1 and CA3 zone in 1 d pretreatment subgroup were significantly increased (P < 0.05-0.01).Conclusion Clonazepam pretreatment can increase the expression level of GABAAR γ2 in the hippocampus of epileptic rats.
