金属硫蛋白对ApoE基因缺陷小鼠血脂、动脉粥样硬化及主动脉SR-AⅠ表达的影响

摘要
Abstract
Objective To observe the effects of metallothionein(MT) on the blood lipids,atherosclerosis and class A Ⅰ scavenger receptors (SR-A Ⅰ) expression in ApoE-deficient (ApoE-/-) mice.Methods A total of 20 six-week-old ApoE-/-male mice were randomly divided into positive control group and MT group.Fatty diet was given for 13 weeks.Another 10 wild male mice were selected as negative control group and normal diet was given for 13 weeks.After 13 weeks,eyeball blood was taken from ApoE-deficient mice to detect TC,TG and LDL-C.The rats were executed to get the aorta and morphology changes of aortic wall were observed after HE staining and SR-A Ⅰ immunohistochemistry staining.The expression of SR-A Ⅰ protein and mRNA were detected by Western Blot and RT-PCR.Results There was no difference in the blood lipid level between MT group and positive control group(P > 0.05)and there was no difference in liver,renal function and creatinine kinase(P >0.05).Compared with the control group,intimal thickness,plaque volume and SR-A Ⅰ protein and mRNA expression increased in positive control group (all P < 0.05).Compared with positive control group,intimal thickness,plaque volume and SR-A Ⅰ protein and mRNA expression decreased in MT group (all P < 0.05).Conclusion MT has no effect on lipid profile,but it can alleviate atherosclerosis lesions and decrease SR-A Ⅰ protein and mRNA expression in aorta of ApoE-/-mice.关键词
