Abstract

Objective To observe the effect of exogenous hydrogen sulfide on the proliferation and apoptosis of human uterine leiomyoma cells and to investigate its mechanism.Methods The human uterine leiomyoma cells were randomly divided into the control and observation group.The cells were treated with different concentrations of sodium bisulfide (NaHS) (10-5 mol/L,10-4 mol/L and 10-3 tmoL/L) for 12,24 and 48 hours in the observation group.MTT assay was used to detect the cell proliferation inhibition rate.The apoptosis rates were measured by flow cytometry.The mRNA and protein expressions of p53 and Bcl-2 in human leimyoma cells were measured by Real-time PCR and Western blot.Results With the increase of NaHS concentration and the prolonged time,the cell proliferation inhibition rates were significantly increased (all P < 0.05) and the apoptosis rates were significantly increased (all P < 0.05) in human uterine leiomyoma cells.In the observation group,with the increase of NaHS concentration and the prolonged time,the mRNA and protein expressions of p53 were up-regulated (all P <0.05),and the mRNA and protein expressions of Bel-2 were down-regulated (all P < 0.05).Conclusions Exogenous hydrogen sulfide inhibits the cell proliferation and promotes the apoptosis of human uterine leiomyoma cells,whose mechanism may be related to the up-regulation of p53 expression and down-regulation of Bcl-2 expression induced by hydrogen sulfide.
