Abstract

Objective:To explore insulin-like growth factor I(IGF-I)levels in serum and follicular fluid and insulin-like growth factor I reeeptor(IGF-IR)expression in granular cells of ovarian cumulus,and analyze the impact of IGF-I and IGF-IR on oocyte maturation of patients in stimulated cycles.Methods:A total of 32 patients received intracytoplasmic sperm injections(ICSI)were enrolled in this study.The samples of blood and follicular fluid as well as granular cells were obtained on the day of oocyte pick-up (OPU).The IGF-I levels in serum and follicular fluid were measured by enzyme-linked immunosorbent assay.The IGF-IR mRNA expression in granular cells was analyzed by real-time PCR.The oocyte maturation was also recorded.Results:The ratio between IGF-I levels in serum and number of oocytes retrieved was positively correlated with IGF-I levels in follicular fluid.The IGF-I levels in follicular fluid of MII period oocytes were significantly higher than those of MI period oocytes(P<0.05).The IGF-IR mRNA in granular cells of MII period oocytes was three times higher than that in MI period oocytes(P<0.05).Conclusions:The IGF-I levels in follicular fluid are positively correlated with the expression of IGF-IR mRNA in granular cells.The interaction of IGF-I and IGF-IR may affect oocyte maturation.The ratio of IGF-I levels in serum and number of oocyte retrieved may have predictive value for maturation of oocyte.
