Abstract
Objective To investigate the effects of ischemic preconditioning (IP) on the expressions of Nogo-A mRNA, Nogo-A and NgR protein after focal cerebral ischemia-reperfusion injury in adult rats. Methods A total of 152 adult male SD rats were randomly divided into three groups: control group (n = 8), non-ischemic preconditioning group (NIP, n = 72) and IP group (n = 72). The latter two groups were randomly divided into day-1, day-3 and day-7 subgroups (24 in each subgroup). Middle cerebral artery occlusion model was induced by intraluminal suture method. After 10 minutes of IP, 2-hour re-ischemia and 1-day reperfusion were performed at day 1, day 3 and day 7. The brain infarct volume was evaluated with 2,3,5-triphenyltetrazolium chloride (TTC). The expressions of Nogo-A mRNA in the three groups were detected by RT-PCR method whereas the positive expressions of Nogo-A and NgR protein in the ischemic cerebral cortex were observed by immunohistochemical method. Results Compared with those in the corresponding NIP subgroups, the neurological deficit score and brain infarct volume were decreased significantly, and the expressions of Nogo-A mRNA, Nogo-A and NgR protein were downregulated significantly in IP day-1, day-3 and day-7 subgroups (all P<0.05). The brain infarct volume as well as Nogo-A mRNA, Nogo-A and NgR protein expressions were decreased most significantly in IP day-3 subgroup (P<0.05), but they did not differ significantly in day-1 and day-7 subgroups (P>0. 05). Conclusion The downregulated Nogo-A and NgR expressions induced by cerebral ischemic preconditioning may play an important role in brain ischemic tolerance.关键词
