Immobilization of Acetylcholinesterase with Chitosan Coated Glass Fibre Membrane
摘要
Abstract
Acetylcholinesterase is immobilized on chitosan coated glass fibre membrane using glutaraldehyde as cross-linking reagent.The benificial immobilizing conditions are obtained as follows:1 mL of enzyme solution is prepared with 0.2 mol/L phosphate buffer(pH 8.0) adding 50 μL 150 U/mL AChE solution,50 μL 5% (V/V)glutaraldehyde and 100 μL 1% (W/W) BSA.The chitosan coated glass fibre membrane is immersed in the enzyme solution for 8 h at 4 ℃.The optimum temperature and pH value of immobilized AChE are 37 ℃ and 8.0,and the immobilized AChE could be reused four times.关键词
