Effect of PXR gene polymorphism on concentration/dose ratios of tacrolimus in kidney transplant patients

Abstract
Objective To investigate the effect of PXR gene polymorphism on the concentration/dose ratios of tacrolimus in kidney transplant patients.Methods Blood samples were collected from kidney transplant patients who received tacrolimus.The PXR genotypes were determined by the polymerase chain reaction (PCR)-direct sequencing.Tacrolimus tough concentration was measured by ELISA.The concentration/dose ratio were collected.Results PXR-C24381A gene polymorphism was significantly related with the concentration/dose ratios of patients.Patients with A/A genotype carrier was significantly lower than those with C/A genotype or C/C genotype (P ＜ 0.05).No difference was found among the PXR-A7635G genotypes (P ＞ 0.05).Conclusion PXR-C24381A polymorphism is correlated with the whole blood concentration/ dose ratio in kidney transplant patients.关键词
