Abstract
Objective To investigate the effects of matrine (Matrine,referred to as Mat) pharmacological activity,and study inhibitory effect of matrine on hyperplastic scar fibroblasts proliferation,provide theoretical support and experimental basis for the treatment of hyperplastic scar after skin injury.Methods In vitro different concentration gradient of Mat were used to cultured hypertrophic scar fibroblasts,then detected A0 value and LDH value of MTT reaction,etected the cell cycle and the relative content of DNA by flow cytometry,before and after medication examined expression of nuclear Proliferating Antigen (proliferatingcell nuclear antigen,PCNA) by immunohistochemistry.Results In a certain range,hypertrophic scar fibroblasts were induced dosedependently proliferation inhibitory,but the LDH value did not change significantly.Relative content of DNA had no obvious change,expression of PCNA decreased.Conclusion Mat could significantly inhibit proliferation of hypertrophic scar fibroblasts without causing cell necrotic changes in vitro,but Mat had no obvious effect on the content of DNA.关键词
