自发性高血压大鼠血清脂联素和心肌脂联素受体1的表达及胰岛素抵抗程度与心室重构的关系研究
Relationship of Expressions of Adiponenctin,Adiponectin Receptors 1 and the Degree of Insulin Resistance to Ventricular Remodeling in Spontaneously Hypertensive Rats

Abstract
Objective To observe the expression of serum adiponectin hormone ( APN ), myocardial adiponectin receptor 1 ( AdipoR1 ), the degree of insulin resistance ( IR ) in spontaneously hypertensive rats ( SHR ), to explore their relationship to elevation of blood pressure and ventricular remodeling. Methods The serum APN, fasting insulin ( FINS ) were determined in 22 - week - old SHR ( SHR group, n = 8 ) and Wistar - Kyoto rats ( WKY group, n = 8 ) . The distribution of myocardial AdipoR1 and its protein expression were detected by immunohistochemical method, AdipoR1 mRNA by real - time PCR. Results As compared with WKY group, BW reduced, SBP, HW, LVW, HWI, LVWI, FINS and HOMA - IR increased in SHR group, the difference was significant ( P < 0. 05 ), but there was not difference in FBG ( P > 0. 05 ) . There was difference in APN expression between 2 groups ( P < 0. 05 ) . The AdipoR1 positive staining decreased in SHR group, differing from WKY group ( P <0. 05 ) . AdipoR1 nRNA expression was lower in SHR group than in WKY group, the difference was significant ( P < 0. 05 ) .By Pearson linear correlation, SBP was negatively correlated with APN , AdipoR1 protein, AdipoR1 mRNA , positively with HWI, LVWI, HOME - IR; By multiple linear regression analysis, AdipoR1 protein, AdipoR1 mRNA, HWI, SBP were influencing factors of LVWI. Conclusion SHR has obvious ventricular remodeling, serum APN and AdipoR1 expression reduce; SHR ventricular remodeling may be correlated with the reduction of serum APN and AdipoR1 expression.关键词
