浙江省甲型H1N1流感病毒血凝素基因序列分析
Sequence analysis of hemagglutinin gene of pandemic influenza A (H1N1) viruses in Zhejiang Province

Abstract
The aim was to analyze the evolutionary characteristics of hemagglutinin gene of pandemic influenza A (H1N1) virus isolated from Zhejiang Province during 2009 and 2011 . Viral RNA was extracted from 8 influenza A (H1N1) virus strains . Hemagglutinin gene was amplified by RT-PCR and the ORF sequence was joined together using sequencing data . Hemagglutinin genes from 20 representative domestic and overseas strains during 2009 to 2012 were downloaded from GenBank database for analysis . All the sequences were used for construction of phylogenetic tree by MEGA software . The 1 701 bp ORF sequences of hemagglutinin gene were obtained by analyzing sequencing results of the 8 strains . After alignment, high homolo-gy were observed among all the HA genes originated all over the world . The homology between 3 strains from Zhejiang Province during 2009 and 2010 and the representative strain of A/California/04/2009 (H1N1) from North America was as high as 99.4% -99.5% . The homology between 5 strains from Zhejiang Province isolated in 2011 and the A/California/04/2009 (H1N1) was 98 .5% -99 .1% . A D222G mutation in hemagglutinin protein was observed in the A /Zhejiang-Yiwu/11/2009 (H1N1) strain isolated from a patient with severe influenza A (H1N1) infection . Although more mutations were accumulated in HA genes in the strains from Zhejiang Province in 2011 comparing with strains from early stage of the pandemic , high homology of more than 98 .5% was observed in all the HA genes . A D222G mutation in HA might be related to the severe illness .关键词
