Abstract

OBJECTIVE:To develop fingerprint ofAtractylodes macrocephala and stir-baked Atractylodes macrocephala with bran,and to compare the difference of fingerprint between them.METHODS:HPLC method was adopted.The determination was performed on Diamonsil C18(250 mm×4.6 mm,5 μm) column with mobile phase consisted of acetonitril-water (gradient elution) at the flow rate of 0.8 ml/min.The detection wavelength was set at 242 nm,and the column temperature was 28 ℃.The analytic time was 100 min.RESULTS:Under the above chromatographic conditions,the mutual mode of HPLC fingerprints was set up for Atractylodes macrocephala; stir-baked Atractylodes macrocephala with bran had a similar fingerprint chromatogram to crude drug (similarity>0.9),but there was a little difference in their concent; among 23 common peaks,No.6,10 and 3 peaks were identified as atractylenolide Ⅰ,Ⅱ,Ⅲ.CONCLUSIONS:Established fingerprint is in good precision,reproducibility and stability,which can be used as evidence for quality evalution ofAtractylodes macrocephala and processed product.
