内质网应激相关分子CHOP和磷酸化JNK在高血压全脑缺血大鼠海马区的表达
Endoplasmic Reticulum Stress-related Molecules CHOP and Phospho-JNK Expressions in Rat Hippocampus after Global Cerebral Ischemia Induced by Hypertensions

Abstract
Objective To observe the expressions of CAAT/enhanced I-binding protein homologous protein (CHOP) and phosphorylated JNK expressions in hippocampus after cerebral ischemia/reperfusion in normal rats and spontaneously hypertensive rats with cerebral ischemia,so as to explore the roles of CHOP and JNK in hypertension aggravated global cerebral ischemia reperfusion injury.Methods 60 male Wistar-Kyoto (WKY) rats were randomly divided into sham operation group (sham operation,SO) and global cerebra ischemia-reperfusion group (ischemia/reperfusion,I/R).Another 30 male spontaneously hypertensive rats were selected as whole brain ischemia-reperfusion group (SHR + I/R).Global cerebral ischemia model was formed by using improved four-vessel occlusion of Pulsinelli's method.After 6,24,48 hours,hematoxylin eosin staining was perform to observe neuronal changes in cell morphology.Immunohistochemical staining and Western blot were performed to detect the expression of CHOP and phosphorylated JNK in hippocampus.Results The density of surviving neurons was markedly lower in I/R group compared with the SO group (P < 0.05);the CHOP and phosphorylated JNK proteins were markedly higher in I/R group compared with the SO group (P < 0.05).The CHOP protein and phosphorylated JNK protein increased and reached peak at 24 h.Compared with the I/R group,the density of surviving neurons in SHR + I/R group were significantly lower,the CHOP proteins were markedly higher at 6 and 24 h while lower at 48 h (P < 0.05),and the phosphorylated JNK proteins were significantly higher at all testing time points (P < 0.05).Conclusion Severe nerve cells loss is observed in hypertensive rats after global cerebral ischemia,which is related to upregulation of phosphorylation of JNK and CHOP.关键词
