Abstract
Objective Whether could human umbilical cord mesenchymal stem cell (HuMSC) differentiate into myoblast.Methods HuMSCs being isolated by adherent culture,checking surface markers by flow cytometry,observation of cells ultrastructural with transmission electron microscopy and atomic force electron microscopy,5-Aza inducting HuMSCs to myoblast differentiation,RT-PCR and Western Blot detecting myoblast specific genes and protein expression respectively.Results Stem cells could isolate from umbilical cord with adherent culture.HuMSCs don 1 express CD31,CD45,CD34,strongly express CD29,CD90,CD44,CD105,and extremely low express HLA-DR,CD40.Expression of myoblast special genes and proteins was significantly higher between experimental group and control group.Conclusion Adherent culture was a reliable and economic method.5-Aza could induce HuMSCs to myoblast differentiation.关键词
