Effect of adhesion to fibronectin via β1 integrin on proliferation and apoptosis in A2780cells

Abstract
Objective:To investigate the effect of adhesion to fibronectin via β1 integrin on proliferation and apoptosis in A2780 cells.Methods:The A2780 cells were cultivated in vitro.Cells of control group and experimental group A were separately planted on poly-lysine and FN coated wells.Cells of group B were blocked by the monoclonal antibody of β1 integrin before planted on FN coated wells.Cell proliferation were measured by CCK-8 assay.The apoptosis and cell cycle were analyzed by flow cytometry.Real-time PCR was used to detect the expression of Aurora B and Survivin gene.Results:Compared to the poly-lysine control,A2780 planted on FN had higher proliferation rate,lower apoptosis rate (P＜0.05),and the percent of cells in S or G2/M and the expressions of Aurora B and Survivin gene were increased (P＜0.05).β1 integrin monoclonal antibody could block the activity induced by cell adhesion to FN via integrin β1 in A2780 (P＜0.05).Conclusion:β1 integrin mediated FN adhesion can increased Aurora B and Survivin gene levels,and that may be related to cell growth and apoptosis inhibition induced by FN adhesion.关键词
